Optimised protocols for RNA extraction from a broad taxonomic range of algae

Abstract Despite advancements in RNA extraction methods, from sources rich polyphenols and polysaccharides such as algae seagrasses remains a challenge. Here we present strategy using hexadecyltrimethylammonium bromide (CTAB) buffer demonstrate its effectiveness on broad range of red, green, brown algae, well the cyanobacterium Arthrospira platensis seagrass Zostera marina . For vast majority tested samples achieved high yields comparable to those obtained higher plants by commercially available kits (ranging 3.9 125.9 µg g −1 fresh weight). Analysis UV/Vis spectrometry capillary electrophoresis revealed purity integrity extracts. highly challenging species like Fucus vesiculosus , serratus Dictyosiphon foeniculaceus established an alternative procedure sodium dodecyl sulfate (SDS) combination with commercial kit. With this protocol, even up 317.0 weight were extracted polysaccharide-rich tissues. This study can serve guideline starting point for development protocols so far unstudied algal very diverse taxa.
